Capsule fill rate measures the share of encapsulated doses that fall inside the approved fill-weight window on a two-piece or liquid-fill encapsulation line. Production supervisors, QA technicians, and validation engineers in dietary supplement manufacturing track it shift by shift because fill weight directly governs label-claim potency and USP weight-variation compliance. A drop in fill rate usually signals a worn dosing disc, segregated blend, or tamping pin wear before scrap numbers spike. Catching it early protects yield, avoids costly reconciliation deviations, and keeps the line inside its validated operating range.

How to use the result
- Use it at the end of each encapsulation run or at AQL sampling intervals to confirm the line is holding its validated fill window.
- It treats every out-of-spec capsule equally, so it will not distinguish a capsule a milligram light from one that is grossly underfilled or empty.

- How do you calculate capsule fill rate? Divide capsules within the fill-weight spec by total capsules filled, then multiply by 100. With 9,650 in-spec out of 10,000, the fill rate is 96.5%.
- What is a good capsule fill rate? Well-run supplement lines typically hold 97-99% on standard powder fills. The 96.5% in our example sits 1.5 points under a 98% target, which warrants a check of dosing disc and blend uniformity.
- Why is my capsule fill rate dropping? Common causes are blend segregation in the hopper, worn tamping pins or dosing discs, low or inconsistent powder bed height, and poor flowability from insufficient lubricant or glidant.
- Is fill rate the same as encapsulation yield? No. Fill rate measures weight-conformance of filled capsules, while overall yield also subtracts joining failures, telescoped or dented capsules, and reconciliation losses across the full run.
- What does the fill rate gap to target tell me? It is the points of improvement needed to hit your acceptance goal. A 1.5-point gap on a 10,000-capsule run equals roughly 150 extra capsules you would need inside spec.
